Long-extinct cousins' genetic mysteries revealed

Ancient DNA was extracted and analyzed using a novel sequencing approach.

Degraded DNA molecules from a group of human relatives who went extinct tens of thousands of years ago have been reassembled using a new technique, yielding a genetic code for the mysterious Denisovans that meets the standard for modern humans.

The findings are based on samples drawn from 40 milligrams of ground-up bone from a Siberian girl's finger. They confirm what scientists saw in a much less detailed genetic sequence they produced a couple of years ago and address some of the deep questions surrounding the Denisovans. But they also raise a few new questions, including a basic one: Just how old was the sample they analyzed?

"The amazing thing is that we can sequence the whole genome to very high accuracy, but there is too little carbon in it to do a date," Svante P??bo, a genetic researcher at the Max Planck Institute for Evolutionary Anthropology, told reporters during a teleconference this week. P??bo is the leader of the research team behind a paper on the project, published online today by the journal Science.


The finger bone was found in southern Siberia's Denisova Cave during an excavation in 2008. That dig was aimed at untangling the genetic relationship between Neanderthals and modern humans, and bones representing both those species were indeed found in the cave. But the initial analysis of the finger-bone sample revealed a genome that was neither Neanderthal nor classically human. That tiny bone and two molars that were also found in the cave represent the only known specimens of the Denisovan race.

Previous analyses of the Denisovan genome were considered merely a rough draft, doing no better than an average of 1.9 readings for every molecular base pair in the DNA, or 1.9X. In comparison, the latest analysis goes to a coverage depth of 30X, which is typical of whole-genome sequencing for present-day humans.

The accuracy was increased by taking short, degraded double-strands of DNA, which couldn't be analyzed ?the usual tools for gene sequencing, and "unzipping" them into single strands. That made it easier to attach specially designed molecules known as adapters and read out the DNA code, piece by piece.

"There are many things you can do with such a high-coverage genome that you cannot do with the low-quality genome that we had before," said Matthias Meyer, a colleague of P??bo's at the Max Planck Institute who developed with the new sequencing technique. Here are the main findings reported today:

  • A comparison of chromosomes that the girl inherited from her mother and father indicated that there was surprisingly low genetic diversity in the Denisovan population. That would suggest that the Denisovan population never was very large, and could explain why the group faded into extinction as populations of modern humans spread out.
  • Detailed study of the "gene flow" over time suggests that the Denisovans interbred with Neanderthals as well as our own species, Homo sapiens. The analysis confirmed previous findings that Denisovans were more genetically similar to Neanderthals than to anatomically modern humans. So does that mean that Denisovans were a separate species? "I really stay away from species designation," P??bo told reporters. "I would not call it a different species, but clearly different groups with a different history. I would not call the Neanderthals a different species from humans either, actually."
  • A comparison of the genome with those of modern-day human populations confirmed that Melanesians, Australian Aborigines and other Southeast Asian islanders had the most in common genetically with the Denisovans. The Denisovan contribution to the genomes of present-day Papuans was estimated at 6 percent.
  • A deeper analysis of the Papuan-Denisovan connection showed that the Denisovan contribution was lower for the sex-linked X chromosome than it was for other chromosomes. That might suggest that the Denisovan males were more likely than the females to interbreed with modern humans. Or it could mean there was a genetic defect on the X chromosome that led to its elimination from modern-day genomes.
  • The researchers were able to triangulate, using the Denisovan genome as well as the genetic codes for Neanderthals and modern humans, to come up with an unexpected result: Present-day east Asians appear to have more in common genetically with the Neanderthals than present-day Europeans, even though Europe was thought to be the main hangout for Neanderthals hundreds of thousands of years ago.
  • The researchers also drew up a catalog of more than 100,000 genetic differences that apparently arose between modern humans and the now-extinct Denisovans and Neanderthals in the past 100,000 years or so. About 260 of the changes affect protein function, P??bo said. "It is quite interesting to me to note that eight of them have to do with brain function and brain development ... and some of them have to do with genes that, for example, cause autism when these genes are mutated," he said. "So I think this is perhaps in the long term, to me, the most fascinating thing about this: what it will tell us in the future about what makes us special in the world, relative to Denisovans and Neanderthals."

Genetic sequencing alone can't tell scientists how long ago the Denisovans lived, however. P??bo and his colleagues factored in assumptions about the mutation rate of the human genome to estimate that the girl with the finger bone lived somewhere around 74,000 to 82,000 years ago. But a separate line of evidence, based on the rock layering in the cave, suggested that the bone was 30,000 to 50,000 years old.

"I'm very unsure about the archaeological dates, but I would say that I am equally unsure about our molecular dates," P??bo told reporters.

Similarly, the researchers give a wide range of dates for their estimate of the time when the Denisovan population split off from the evolutionary line leading to modern humans: 172,000 to 700,000 years ago.

"Most of the uncertainty in that number ... comes from the uncertainty at present about the human mutation rate," said another co-author of the paper, David Reich, a geneticist at Harvard Medical School and the Broad Institute. "There is a lot of debate in the community about the rate at which mutations accumulate."

Future studies, involving DNA analysis as well as carbon-dating analysis of other specimens from the Denisova Cave, may help clear up that uncertainty. But for the team behind the research announced today, an even higher priority is to generate a gene sequence for the Neanderthals that's as good as the sequence they now have for the once-mysterious Denisovans.

John Hawks, an anthropologist at the University of Wisconsin at Madison, said the newly published paper was "solid work."

"It's really neat that they've organized a catalog of things that humans have that are not present in this ancient genome," Hawks, who studies human origins but was not part of P??bo's research team, told me. He said the study of comparative brain genetics, for example, was becoming an increasingly important area, "and this study is part of it."

"We know that the human brain is a big target of evolution, but establishing the order of these changes is an important step now," he said. "This gives us a time stamp on some of those changes."

The technique developed for the Denisovan DNA could be applied much more widely in the future. Meyer said his method could be used on modern-day forensic samples that are too degraded to be analyzed using current techniques. And who knows? Anthropologists may find still more long-lost, extinct cousins in the fossil record whose genomes need to be done.

Scientists have established a genetic mouse model for primary ovarian insufficiency (POI), a human condition in which women experience irregular menstrual cycles and reduced fertility, and early exposure to estrogen deficiency.

POI affects approximately one in a hundred women. In most cases of primary ovarian insufficiency, the cause is mysterious, although genetics is known to play a causative role. There are no treatments designed to help preserve fertility. Some women with POI retain some ovarian function and a fraction (5-10 percent) have children after receiving the diagnosis.

Having a mouse model could accelerate research on the causes and mechanisms of POI, and could eventually lead to treatments, says Peng Jin, PhD, associate professor of human genetics at Emory University School of Medicine.

The results were published online recently in the journal Human Molecular Genetics.

The paper was the result of a collaboration between researchers at Emory and the Institute of Zoology, Chinese Academy of Sciences in Beijing. Dahua Chen, PhD, associate director of the State Key Laboratory of Reproductive Biology, is the senior author and postdoctoral fellow Cuiling Lu is the first author. Stephanie Sherman, PhD, professor of human genetics at Emory, is a co-author.

The mouse model builds on research on women who are carriers of a "premutation" for fragile X syndrome, a leading cause of inherited intellectual disability.

The mice have a fragment of a human X chromosome from a fragile X premutation carrier. Other non-genetic mouse models used to study menopause include surgical removal of the ovaries, or exposure of mice to a chemical, 4-vinylcyclohexene diepoxide, which depletes the ovaries.

"While the fragile X premutation is a leading cause of POI, I think this model will be useful and relevant for all women with this condition," Jin says.

Women with the fragile X premutation account for around two percent of spontaneous POI cases and 14 percent of familial POI cases. About 20 percent of women who carry the fragile X premutation experience POI, the disorder now called fragile X-associated POI, or FXPOI.

Fragile X syndrome is caused by the expansion of a "triplet repeat" in a gene (FMR1) that is important for signaling in the brain. In fragile X syndrome, the triplet repeat -- three DNA letters (CGG) repeated many times -- forces the gene to shut off.

For a woman who carries the premutation, the triplet repeat is not large enough to shut the gene off. There is a risk that it will expand in her children enough to generate fragile X syndrome. In addition, the triplet repeat appears to have an effect on the woman's ovaries, independently from its influence on the FMR1 gene.

Jin says studying mice that have an analogous genetic alteration will help scientists understand what's happening to the ovaries in POI. It appears that the RNA coming from the premutation impairs development of the ovarian follicles, the structures in which eggs/oocytes mature.

The research team found that a quarter of premutation-carrying female mice are infertile. When they are housed with male mice, those that do have pups have them a month later on average (12.5 weeks of age compared to 8.5 weeks), and they have fewer pups.

Puberty occurs at roughly five weeks of age in mice, and the premutation mice have alterations in their ovaries already before puberty. At 25 days of age, there are a reduced number of mature follicles in ovaries of the female mice carrying the premutation. Those mice also have altered levels of hormones resembling those of women with POI, such as elevated FSH (follicle stimulating hormone).

The research team found that in the ovaries of mice with the fragile X premutation, ovulation-related genes are less active. In addition, two cellular signaling pathways (Akt/mTOR) are less active in the ovaries, suggesting that drugs that affect those pathways could be used to treat POI.

Kepler discovers planetary system orbiting 2 suns

Astronomers at the International Astronomical Union meeting announced the discovery of the first transiting circumbinary multi-planet system: two planets orbiting around a pair of stars. The discovery shows that planetary systems can form and survive even in the chaotic environment around a binary star. And such planets can exist in the habitable zone of their stars. "Each planet transits over the primary star, giving unambiguous evidence that the planets are real," said Jerome Orosz, Associate Professor of Astronomy at San Diego State University and lead author of the study which is published today in the journal Science.

The system, known as Kepler-47, contains a pair of stars whirling around each other every 7.5 days. One star is similar to the Sun while the other is a diminutive star only one third the size and 175 times fainter. The inner planet is only 3x larger in diameter than the Earth, making it the smallest known transiting circumbinary planet. It orbits the stellar pair every 49 days.

The outer planet is slightly larger than Uranus and orbits every 303 days, making it the longest-period transiting planet currently known. More importantly, its orbit puts it in the "habitable zone", the region around a star where a terrestrial planet could have liquid water on its surface. While the planet is probably a gas-giant planet and thus not suitable for life, its discovery establishes that circumbinary planets can, and do, exist in habitable zones.

Although much more difficult to detect than planets around single stars, the rich dynamics and wild climate changes make these circumbinary planets worth the effort to find. These two planets join the elite group of 4 previously known transiting circumbinary planets, Kepler-16, 34, 35 and 38.

The new planetary system is located roughly 5000 light-years away, in the constellation Cygnus. The planets are much too far away to see, so they were discovered by the drop in brightness they cause when they transit (eclipse) their host stars. The loss of light caused by the silhouette is tiny, only 0.08% for planet b and 0.2% for planet c. By comparison, Venus blocked about 0.1% of the Sun's surface during its recent transit. Precise photometric data from NASA's Kepler space telescope allowed the transits and eclipses to be measured, which in turn provided the relative sizes of the objects. Spectroscopic data from telescopes at McDonald Observatory in Texas enabled the absolute sizes to be determined. "Based on their radii, these probably have masses of approximately 8 and 20 times that of the Earth," Orosz said.

"Kepler-47 shows us that typical planetary architectures, with multiple planets in co-planar orbits, can form around two stars," said co-author Joshua Carter, a Hubble Fellow at the Harvard-Smithsonian Center for Astrophysics. "We've learned that circumbinary planets can be like the planets in our own Solar System, but with two suns."

The work was presented at the International Astronomical Union meeting by Dr. William Welsh, Professor of Astronomy at San Diego State University, on behalf of the Kepler Science Team.

"The thing I find most exciting," said Welsh, "is the potential for habitability in a circumbinary system. Kepler-47c is not likely to harbor life, but if it had large moons, those would be very interesting worlds."

The first song ever played from Mars was broadcast today (Aug. 28) from NASA's Curiosity rover on the Red Planet.

The concert represented the world premiere of the appropriately named song "Reach for the Stars," by rapper and songwriter will.i.am. It was beamed via radio signal about 150 million miles (241 million km) back to Earth from Curiosity's landing spot on Mars' Gale Crater at 1 p.m. PDT (4 p.m. EDT).

"The point of the song is to remind people ? that anything is possible if you discipline yourself and dedicate yourself and stand for something," the musician said today from Curiosity's mission control center at NASA's?Jet Propulsion Laboratory?(JPL) in Pasadena, Calif.

The music arrived there during an educational event held for students from the Los Angeles neighborhood where will.i.am grew up.

"There's no words to explain how amazing this is," will.i.am. said. "These kids here are from Boyle Heights, the same neighborhood I'm from. We don't have to just end up in the 'hood. But it's a hard thing. The hardest thing is discipline."

Students and rocket scientists alike bobbed their heads and clapped as the song's strains came back to Earth.

"Why do they say the sky is the limit
When I've seen footprints on the moon
And I know the sky may be high
But baby it ain't really that high
And I know that Mars might be far
But baby it ain't really that far
Let's reach for the stars"

Also onhand at the event was former NASA astronaut Leland Melvin, now the agency's associate administrator for education.

"Never give up," Melvin said. "People told me that I couldn't be an astronaut. Whatever you want to be, whatever you dream, you can do, if you put your mind to it."

The $2.5 billion Curiosity landed on Mars Aug. 5 (PDT) after launching in November of last year. The song was loaded onto the rover before its departure from Earth, and will.i.am, part of the hip-hop group The Black-Eyed Peas, was at JPL in person to?watch Curiosity land.

The car-size rover is embarking on a two-year mission to search for signs that the Red Planet may have ever been?habitable to microbial life.

PERRY HALL, Md. - A 15-year-old sophomore at a suburban Baltimore high school who made references to murder-suicide on Facebook has been charged as an adult in the shooting of a classmate on the first day of school, authorities said Tuesday.

Robert Wayne Gladden Jr. was being held without bail on charges of attempted first-degree murder and first-degree assault, Baltimore County police said. A preliminary hearing was scheduled for Sept. 7. The state's attorney's office said it?did not know if he had a lawyer.

Gladden's last status update on his Facebook page, posted the morning of the shooting, read: "First day of school, last day of my life. ... f--- the world."

A 15-year-old Maryland gunman was charged as an adult after he shot and critically wounded another student at their local high school. WRC's Pat Collins reports.

His father told The Associated Press that his son had been bullied. Baltimore County Police Chief James Johnson said at a news conference Tuesday that he was aware of the reported bullying, but he said Gladden has not indicated in conversations with detectives that bullying was a motive for the shooting. His father did not disclose other possible motives.

Gladden continues to cooperate with investigators and was undergoing a mental health evaluation, Johnson said.

Gladden rode to school on the bus Monday morning with a bag containing a disassembled shotgun, 21 rounds of ammunition and a bottle of vodka, Johnson said.

When he arrived at school, Gladden went to his first two classes, Johnson said. On the way to the cafeteria, he stashed the bag with the shotgun in a restroom, the chief said. A short time later, he returned to the restroom and assembled the gun, which he then hid beneath his clothes, Johnson said.

Upon entering the cafeteria, he pulled out the gun and fired a shot toward a lunch table, according to charging documents. A 17-year-old classmate, Daniel Borowy, was struck in the back.

Borowy is a special needs student, according to NBC affiliate WBALTV.com.?He remained in critical condition Tuesday at Maryland Shock Trauma Center, a hospital spokeswoman said. His family issued a statement asking well-wishers to keep him in their thoughts and prayers and asking for privacy.

Teachers and school staff rushed toward Gladden, and in the ensuing struggle, he fired another shot that hit the ceiling, investigators said in the documents. The staffers were able to get the gun away from him and he was arrested by a school resource officer.

Witnesses credited guidance counselor Jesse Wasmer with getting the gun away from Gladden.

Gladden sipped from the vodka bottle before the shooting but did not drink enough to become intoxicated, Johnson said.

The teen got the shotgun from his father's house, Johnson said. The weapon was manufactured before 1968 and was of legal length, and police were trying to determine whether it was properly registered, he said.

On his Facebook page, Gladden referred to mass murderer Charles Manson and gave himself the nickname "SuicidalSmile." The page, identified by classmates as belonging to the suspected shooter, was just launched in July, and the three photos of Gladden all show his face hidden behind long, dark hair. He describes himself as a "metalhead" and a fan of musicians Marilyn Manson and Slipknot.

The suspect's father, Robert W. Gladden, told the AP Monday evening that his son was the shooter and indicated his son had been bullied. He gave no further details.

A woman who was also at the home in Middle River and said she was related to the elder Gladden gave the following statement on the family's behalf: "We are horrified. We did not see this coming and our thoughts and prayers are with the victim and the victim's family."

Court documents show Gladden's parents divorced in 2010, reported WBALTV.com. The news station?went to the house where Gladden lives with his mother on Monday afternoon; a sign on the door read "We don't call 911," and had a picture of a revolver on it, reporters said.??No one in the house would comment on the shooting.

Police also executed a search warrant at the Kingsville home of Gladden's mother and stepfather and arrested the stepfather ? Andrew Piper, 43 ? on illegal gun and drug possession charges, police said. The charges against Piper had nothing to do with the school shooting, Johnson said, but he noted that Gladden lived part-time at that address.

Classes resumed Tuesday at the school amid a low-key police presence. About 150 students turned out for a prayer vigil organized by local churches on the school grounds. Some students wore T-shirts and bracelets reading "Pray for Daniel" and "Team Wasmer" in reference to the victim and the guidance counselor.

Shane Boyer, 44, who was dropping off his 13-year-old daughter Corinne, said the vigil helped calm students still dealing with the shooting. Boyer said he knew immediately that his daughter was safe after the shooting because he received text messages from her. One read, "Someone just shot a gun in lunch," followed by another that said, "I ran out I am do (sic) scared there's helicopters + cops + we are all outside."

Perry Hall is a middle-class community along the Interstate 95 corridor, northeast of the city of?Baltimore. The school is the largest in the county, with 2,200 students.

My friends, Paul and Susie, are about to have there second child, in fact the baby due date is tomorrow (Monday 27th August). I haven?t seen them for a while and so decided to visit them in Milton Keynes, an area known for it?s plastic cows, actually they are concrete as I was forcibly reminded (lol hahaha) and lots of roundabouts. When I arrived we went to Mortimer Square which is an area surrounded by restaurants, most of them chains, like Caf? Rouge, Las Iguanas. We were trying decide where to go but as Susie was almost due it was decided not to have anything spicy, just in case. After looking around for a few minutes it was decided to go to The Turtle Bay, a Caribbean restaurant, odd choice, seeing as Caribbean is renowned for being spicy, but I wasn?t one to argue as I love spicy food.

We entered the large restaurant which was surrounded by large windows and because it was sunny the restaurant had warm and bright feel. In the centre is a large style wooden shack bar which all the tables surrounded. The tables and chairs are all wooden which add to the rustic feel, but it does have a feel of a restaurant chain in the making. When I checked out there website I saw they have another restaurant in Southampton, so I was right a new chain in the making.

Salt Cod Fritters

The menu is extensive with Cutter (starters), Lunch and Lighter (lunch menu or lighter bites if you don?t want a full meal. One Pots, (dishes that comes in a pot ? bet you didn?t see that coming). And Jerk Pit BBQ (dishes from a BBQ with Caribbean flavours). They also have a large array of both alcoholic and non-alcoholic cocktails which are on an almost continuous happy hour of 12-7pm and 10pm-close every day.

Jerk Chicken Wings

I guess they don?t see themselves as a family restaurant as they don?t have a child?s? menu. If you do take a young child in with you it appears that if you don?t want spicy food for your child you need to order a meat dish and ask for the spicy not to be added. We ordered a duck wrap, for little Henry and asked for no spice as advised, but when it arrived it was still fairly spicy..

Jerk Chicken Roti

Myself and Susie order fritters, I ordered the salt cod fritters and she ordered the sweetcorn fritter. They were not what we were expecting, they were more like savoury doughnuts rather than fritters. Also my fritters had virtually no salt cod in them and in fact I pulled them apart and only found one very small piece in each of the four fritters. The sweetcorn had more flavour but again more doughnut than fritter. Paul ordered the jerk pit chicken wings, which were very tangy and with a kick, the meat was moist and well cook and I certainly had food envy.

Sweet Potatoe Fries

After a disappointing starter and a slow removal of out plates and a far wait for our main courses our mains were delivered and we were not disappointed. I ordered the Trini Fish Curry, which was salmon, prawns, mussels, squid and white fish in a gorgeous curry spiced coconut milk. The rice and peas were well cooked and complimented the curry wonderfully. Paul ordered the Blue Mountain Curried Goat was also very tasty, the meat was tender and the curry sauce full of flavour, but with one little moan of not enough sauce, but fully enjoyed. Susie ordered Jerk Chicken Roti, which was pulled jerk chicken in a flat bread wrap which was moist with a hot kick. But not over spicy so the baby did not kick in objection or even worse, decide to show his/her face :-)

Curried Goat with Rice & Peas

We also had some side dishes, which I didn?t mind paying extra for as the main courses were reasonably priced. We ordered fried plantain, which were well cooked with lots of sweet flavour. We also ordered Sweet Potatoes Fries, there were delicious if a little greasy, but one of the highlights for me, seeing as I didn?t think sweet potato would work as a ?chip?, but it?s nice to be proved wrong.

Now I don?t normally review drinks, but as it was a cocktail bar and as Susie couldn?t drink, Paul and I decided we should try as many cocktails for her. So that she knew which cocktail tasted the best and to order once the baby was born. Well it was the only excuse we could come up with to allow us to try as many as we could. We tried the classic Long Island Ice Tea, which was the best. Jamaican Mule, okay, but the ginger beer wasn?t strong enough. Elderflower Collins which was fresh with lemon juice and club soda. Mojito, which had far to much ice so at the end we were drinking cold water and we finished off which Tropical Rum Punch which was okay, but we didn?t like the allspice syrup, which I thought would be nice with rum, but for us didn?t work.

The staff were very friendly and polite and couldn?t really fault them. But if I was to grumble, which means I will, they were a little too over enthusiastic. When we sat down, one waiter took our drinks order and then before the drinks came three other members of staff came over to take our drinks order. It was like that for the rest of the meal, but then when we wanted them there were hard to track down. I wonder if I am moaning for the sake of it, because as I said they were friendly and were very good with baby Henry.

For a small chain, I believe the Turtle Bay has a lot going for it and hopefully they will be a great success. I just hope they don?t go like other chains that as they become popular they expand too quickly and they lose there charm and the food starts to really suffer. I can think of two chains that at the beginning were very good, but I wouldn?t go near now as the food has really suffered. So fingers crossed for them and I would love to see one in London.

record seven Tour de France wins and handed a lifetime ban by

the United States Anti-Doping Agency on Friday, but he remained

defiant as supporters rallied around the American cyclist.

Saying that "enough is enough," Armstrong issued a statement

late on Thursday indicating that would not challenge USADA

charges he had doped throughout his career, although he

continued to deny he had ever used performance-enhancing drugs.

While the USADA can remove Armstrong's titles, such a

decision could ultimately rest with the Court of Arbitration for

Sport in Lausanne, Switzerland, should the International Cycling

Union challenge the USADA's ruling.

But weary from years of denial, legal battles and skirmishes

with former teammates and anti-doping chiefs, Armstrong said it

was a fight he no longer had the stomach for.

"Today I will turn the page," Armstrong said. "I will no

longer address this issue regardless of the circumstances."

Less than 24 hours after issuing that statement, Armstrong

was planning 'business as usual' with a weekend bike ride in

Colorado, tweeting: "Excited to be racing the #poweroffour

tomorrow here in @AspenCO. 9000 vert in just 36 miles!"

Armstrong may have turned the page but the story is far from

over.

One of the sporting world's most polarizing figures,

Armstrong remains a hero to millions of cancer survivors for

beating the disease and coming back to win the Tour de France

seven times. To others, he is a drug cheat and fraud.

World Anti-doping Agency chief John Fahey said Armstrong's

decision not to contest the allegations added up to nothing more

than an admission of guilt.

"He had the right to rip up those charges, but he elected

not to. Therefore the only interpretation in these circumstances

is that there was substance in those charges," Fahey told

Reuters in a telephone interview on Friday.

Bill Stapleton, Armstrong's agent for 17 years, strongly

disagreed.

"This is not an admission of guilt, this is a rebuke of a

process that is disgusting and unfair," Stapleton said. "We

believe they're completely out of bounds.

"We've been clear that we don't think they (USADA) have the

authority to do that and we'll see what the people who run the

Tour de France and the International Federation of Cycling have

to say about that."

The debate over Armstrong's guilt will now rage, with some

heavy hitters like longtime sponsor Nike, the world's biggest

sportswear maker, lining up alongside the disgraced cyclist,

while anti-doping crusaders claim victory.

Since 2004, Nike has helped Livestrong, Armstrong's

organization to help cancer survivors, raise over $100 million

for cancer research and created the Livestrong yellow wristbands

that became a global phenomenon with over 84 million bands

distributed.

CONTROVERSIAL CYCLIST

"Lance has stated his innocence and has been unwavering on

this position. Nike plans to continue to support Lance and the

Lance Armstrong Foundation, a foundation that Lance created to

serve cancer survivors," the company said in a statement.

Armstrong, 40, has been one of the most successful and

controversial cyclists of all time, returning to the sport after

beating cancer to win the Tour de France seven straight times,

from 1999 to 2005.

Livestrong takes its inspiration from his achievements and

recovery from testicular cancer that made him a hero to many and

boosted the sport's popularity in the United States.

The Lance Armstrong Foundation, which raised $51 million in

2011, said it received 400 donations totaling $75,000 on Friday,

20 times more than the amount donated on Thursday.

Armstrong also made many enemies throughout his career, with

several of his former teammates and colleagues allegedly ready

to testify that he doped.

Former teammate and deposed Tour de France winner Floyd

Landis accused Armstrong in 2010 of using performance-enhancing

drugs and teaching others how to avoid being caught.

In a bizarre coincidence of timing, Landis appeared in

federal court in San Diego, California, on Friday and entered a

deferred prosecution agreement in which he admitted defrauding

1,765 individuals who donated money to the Floyd Fairness Fund.

The cyclist agreed to pay $478,354 in restitution.

Armstrong also has his loyalists, such as Jim Ochowicz,

director of the BMC cycling team and a longtime friend who

helped him when he was an amateur rider and young professional.

"As a friend of Lance's, I support his decision to call it

an end," said Ochowicz. "He has done so much for our sport over

the years, and I am sad at what has transpired.

"I love him. I know he still has a big fight ahead of him

and his battle of trying to find a cure for cancer and help

survivors and carry on with the Lance Armstrong foundation.

"I think he has earned every victory he's had," he said.

The USADA believes, however, it has enough compelling

evidence to prove Armstrong did not claim his victories fairly.

A quasi-governmental agency created by the U.S. Congress in

2000, the USADA formally charged Armstrong in June with doping

and taking part in a conspiracy with members of his championship

teams.

EVIDENCE AGAINST ARMSTRONG IN HAND

The agency said in a letter to Armstrong that it had blood

samples from 2009 and 2010 that were "fully consistent" with

doping.

Michael McCann, an expert in sports law at Vermont Law

School, said Armstrong's decision not to contest the USADA

charges in arbitration might have been the cyclist's best option

in the face of mounting circumstantial evidence.

"This gives his supporters reason to support him," McCann

told Reuters. "If he had gone to arbitration and lost - which I

think almost definitely would have happened - from a public

relations standpoint, that would have been much more harmful."

In losing his titles, Armstrong joins Canadian Ben Johnson

and American Marion Jones as the highest-profile athletes to

lose championships as a result of doping sanctions.

Johnson was stripped of the 1988 Seoul Olympics 100 metres

title after testing positive for a steroid. while Jones lost her

2000 Sydney Olympics 100, 200 and 4x400 metres relay gold medals

when she confessed she had been taking drugs at the time.

Armstrong faces the possibility of legal action from

promoters and race organizers looking to recover prize money.

It also remains to be seen what impact the USADA's action

will have on his endorsement potential and ability to keep

raising funds for his foundation.

Despite having his reputation tarnished, Armstrong's triumph

over cancer and fundraising efforts make him a valuable

pitchman, marketing experts said.

While Nike was quick to stand by Armstrong, his other

sponsors, including RadioShack, exercise bike maker

Johnson Health Tech, sunglass maker Oakley, owned by

Italy's Luxottica, and Michelob, made by Anheuser-Busch

InBev, have not leapt to his defence.

Robert Boland, professor of sports management at New York

University, said Armstrong's extensive work on cancer would

help blunt the impact of the lost titles.

"His story has not been diminished. Here's a guy who

essentially was at death's door with cancer and came back. That

example still makes him very compelling," he said.

Still, Boland said it would be difficult for Armstrong to

continue to endorse bicycles or bicycle equipment, since he is

now banned from competitions.

"If he can't show up at certain events, how do you use him?"

With the possibility each of his seven Tour de France crowns

could go to other riders, Armstrong maintained that no matter

what the USADA had ruled, those he competed with would always

know he was the true winner.

Spain's Fernando Escartin, who will rise to second from

third in the 1999 Tour de France after the USADA's decision,

said the American would always be the champion.

"For me, Lance Armstrong remains the 1999 Tour winner,

second (Alex) Zulle and third, me," the now-retired Escartin

told Reuters at the Vuelta a Espana race on Friday.

"It's 13 years now since this all happened. It seems

completely illogical and unreal. I don't want to even think

about it."
